Pacific Biosciences is looking for a scientist to help improve the biochemistry of its sequencing consumables. The successful candidate will work within a highly multidisciplinary R&D environment to develop novel biochemical assays on both the ensemble and single molecule levels in order to characterize the interactions between fluorescently labeled nucleotide analogs and engineered DNA/RNA polymerases. Other areas of focus will be to improve nucleic acid sample preparation and subsequent sequencing performance as it pertains to polymerase processivity, DNA/RNA integrity, and secondary structure. Ideally, the candidate will have a strong background in biochemistry, chemical biology and/or biophysics with a good working knowledge of chemistry and quantitative data analysis methods. Direct experience with fluorescence spectroscopy, transient state kinetics, and/or single molecule kinetics with nucleic acid-relevant enzymes is a significant plus.  

•    Ph.D. in Biochemistry, Chemical Biology, or Biophysics
•    Strong background in the kinetic and thermodynamic properties of proteins and nucleic acids 
•    Extensive experience with quantitative in vitro assay design and execution
•    Familiar with various protein labeling and purification techniques
•    Hands-on experience with fluorescence spectroscopy (FCS, FRET, anisotropy, stopped-flow, quench-flow, gel electrophoresis and/or plate-based measurements) 
•    Able to analyze and interpret large and complex data sets (non-linear regression, global fitting, and/or statistical analyses) 
•    Proficient in the use of standard molecular biology techniques (DNA sequencing, PCR, cloning, and mutagenesis)
•    A desire to work at the bench at least 50% of the time
•    Strong organizational and communication skills and the abilities to work within a team and thrive in a fast-paced environment
•    All levels of experience will be considered
